چکیده | ||
زمینه مطالعاتی: باجداسازی فراکسیونهای فنولیآزاد و باندشده و تعیین محتوای فنولی و فعالیت آنتیاکسیدانی آنها میتوان بهعنوان جایگزینی برای آنتیاکسیدانهای سنتزی و یک روش پیشگیری برای کاهش اثرات منفی ناشی از استرس اکسیداتیو استفاده نمود. هدف: این مطالعه باهدف استخراج ترکیبات فنولی و فلاونوئیدی از برگ گیاه زوفا (Hyssopus officinalis) انجام شد. روشکار: با استفاده از اولتراسوند پروب تحت شرایط زمانی ۱۰ و ۲۰ دقیقه و شدت ۶۰ و ۱۰۰ وات و استفاده از حلال اتانول: آب با نسبتهای (۸۰:۲۰) و (۵۰:۵۰) و جداسازی فراکسیون فنولیآزاد از عصارهها با استفاده از حلال متانول و اولتراسوند و سپس جداسازی فراکسیونهای فنولیباندشده با استفاده از حلال سود و اولتراسوند انجامگرفته است. و فعالیت آنتیاکسیدانی توسط آزمونهای مهار رادیکال آزاد (DPPH) و سنجش بیرنگ شدن بتاکاروتن – لینولئیکاسید و آزمون پایداری اکسایشی (OSI)، صورت گرفت. نتایج: تجزیهوتحلیل آماری نشان داد که شرایط بهینه برای استخراج، مربوط به تیمار 80 درصد اتانولی شدت 100 و زمان 20 دقیقه (۸۰-۱۰۰-۲۰) بوده که بیشترین میزان ترکیبات فنولی و فلاونوئیدی و فنولیآزاد و باند شده را دارد و بهترین فعالیت آنتیاکسیدانی و پایداری اکسایشی را فراکسیونهای فنولیآزاد نشان دادند. نتیجهگیری نهایی: میتوان نتیجه گرفت که عصاره برگ زوفا به دلیل مقادیر قابلتوجه ترکیبات فنولی و فلاونوئیدی دارای قدرت آنتیاکسیدانی بالا است که بین این ترکیبات و فعالیت آنتیاکسیدانی رابطه مستقیم وجود دارد و فعالیت آنتیاکسیدانی فنولآزاد بیشتر از فنولباندشده است. همچنین اولتراسوند پروب میتواند یک روش مناسب برای تولید عصارههای گیاهی با فعالیت آنتیاکسیدانی بالا باشد. اولتراسوند پروب، فعالیت آنتیاکسیدانی، زوفا، ترکیبات فنولیک، فنول آزاد، فنول باند شده | ||
